Androstenedione, a weak precursor androgen of adrenal and gonadal origin, is converted to testosterone by 17-beta-hydroxysteroid dehydrogenase. Testosterone is reduced to dihydrotestosterone by 5-alpha-reductase in target tissues. Both androstenedione and testosterone can exit the pathway to estrogens, estrone and estradiol, via aromatase. Dihydrotestosterone is the highest-affinity androgen-receptor ligand and is non-aromatizable.
